포도 과피 및 종자의 탄닌 분석을 위한 추출 용매 조건 탐색

Abstract

The total tannin and individual condensed tannin concentrations in the fruit skin and seeds of “Campbell Early” grapes were analyzed by ultrasound-assisted extraction using various solvents (ethanol, methanol, and acetone, both pure and diluted to 70% with and without 0.5% HCl). The total tannin contents of the grape skin and seeds depended on the extraction-solvent conditions and varied between 0.01 and 2.92 mg/g for the skin, and 8.82 and 14.72 mg/g for the seeds. The most effective solvent for extracting total tannins from both the grape skin and seeds was found to be 70% acetone, which facilitated the detection of high concentrations of tannin monomers and dimers. Monomer tannins (catechin and gallocatechin) concentrations in grape seeds were detected at high levels using both 70% acetone and 70% methanol as the extraction solvents; however, dimer tannins (procyanidin B1 and procyanidin B2) were detected at high levels in grape seeds using 70% methanol. Hence, 70% methanol was effective for measuring the concentrations of condensed tannins in grape seeds. Since we are currently analyzing individual tannins, research into finding the analysis conditions most appropriate for individual situations is still required.

서 론

탄닌(condensed tannin)은 포도의 중요한 폴리페놀 화합물로 과피 및 종자에 존재하고 부위에 따라 함유된 정도가 다르다(1,2). Flavan-3-ol 단일 물질 및 중합체 형태로 proanthocyanidin으로도 불리며 포도 과피와 종자에 위치하며 떫음(astringency)과 씁슬함(bitterness)을 나타내는 요인으로 포도 및 와인의 색과 품질 결정에 크게 관여하는 것으로 알려져 있다(3-5).

Flavan-3-ols은 catechin, epicatechin, gallocatechin, epigallocatechin 및 galloyl기가 결합된 epicatechin gallate 등을 포함하며, 중합 반응을 거쳐 monomer 탄닌이 2~8개 결합한 올리고머(oligomer) 화합물과 10개 이상이 결합한 폴리머(polymer) 화합물을 형성한다(6). 포도의 떫음 정도는 탄닌 농도와 올리고머의 크기와 관련이 있으며 galloyl기가 결합된 gallolation 탄닌에서 높았고, 씁슬함 정도는 폴리머 화합물보다 monomer 탄닌에서 높게 나타났다(3-5,7).

탄닌의 추출 방법으로는 Solid-liquid extraction(SLE), supercritical fluid extraction(SFE), pressurized water extration(PWE), microwave assisted extraction(MAE) 및 ultrasound assisted extraction(UAE) 등이 있으며, 추출 정도는 포도 품종이나 환경적 요인(날씨, 숙성 시기, 토양 조건 및 재배 지역) 등에 따라 많은 차이를 나타낸다(6,8). 그 중에서 초음파 추출법은 식물 세포벽을 붕괴시켜 조직 내로 용매를 침투시킴으로써 세포 내에 존재하는 생리활성물질 추출이 용이하다고 알려져 있으며 다른 추출법에 비해 효율이 6-35% 정도 증가하는 것으로 보고된 바 있다(9,10). 또한, 최근에는 정량 분석을 위한 전처리 방법으로 적은 용매를 사용하여 분석 시간을 줄이면서 분석 성분의 손실은 최소화하고, 폐용매의 발생량을 줄이는 친환경적 분석법이 많이 시행되고 있는데 초음파 추출은 적은 양의 유기용매를 사용하고 초음파 처리를 통한 물질 추출로 추가적인 용매의 사용을 줄일 수 있다는 점과 용매를 생산할 때 발생하는 용매 부산물 및 실험 후 발생하는 폐 용매의 양을 줄일 수 있다는 점에서 친환경적인 분석법이라 할 수 있다(11,12). 그동안 많은 선행 연구에서 methanol, ethanol 및 acetone 등의 추출 용매를 사용하여 탄닌 함량을 측정한 바 있으나, 용매 종류와 추출 부위가 각기 달라 직접적인 비교는 불가능하였다(13-15).

최근의 생식용 포도 품종 개발은 소비자의 기호를 반영한 ‘씨가 없고 껍질째 먹는 아삭아삭한 포도’ 및 떫은 맛과 씁씁한 맛이 없는 품종을 목표로 하고 있다. 품종을 개발하기 위해 육성된 수많은 교배실생의 과실을 직접 먹어보면서 떫은 맛이나 씁쓸한 맛이 나는 실생묘목을 도태한다는 것은 인적 및 시간적 제약이 있으며 정확한 판단이 어렵다. 따라서 포도의 떫은 맛과 씁쓸한 맛이 나지 않는 품종을 개발하기 위해서는 실험실 조건에서 품종의 탄닌 함량 측정을 통해 품종을 선발할 수 있는 탄닌 추출법의 확립이 필요하다. 따라서 본 연구에서는 초음파 추출법을 사용하여 methanol, ethanol 및 acetone에 각각 100%, 70% 및 0.5% HCl를 포함한 70% 용매 조건에서 포도의 과피 및 종자에 존재하는 monomer 및 dimer 탄닌 추출을 최적화 할 수 있는 용매를 탐색하고자 하였다.

재료 및 방법

실험 재료 및 시약

포도는 국내에서 가장 많이 재배되고 있는 Campbell Early 품종을 대상으로, 전북 완주군 이서면에 위치한 국립원예특작과학원 재배포장에서 2018년 수확한 포도를 사용하였다. 정성 및 정량 분석을 위해 실험용 표준물질 catechin, epicatechin, gallocatechin, epigallocatechin, procyanidin B1, B2는 Sigma-Aldrich(St. Louis, Missouri, USA)에서 구입하여 사용하였다. 또한, 탄닌 추출 시 사용된 methanol, ethanol 및 acetone은 순도 98% 이상으로 사용하였으며, high pressure liquid chromatography(HPLC) 분석에 사용한 water, methanol, acetonitrile 용매 또한 Sigma-Aldrich에서 구입한 HPLC grade로 사용하였다.

탄닌 화합물 추출

탄닌 화합물 추출은 Downey와 Hanlin(15)의 추출법을 인용하여 수행하였다. Campbell Early 품종은 재배 포장에서 수확한 후 과피와 종자를 송이에서 조심스럽게 분리하여 각각 거즈 및 증류수로 이물질을 제거하고 실험 전까지 -20℃에서 냉동 보관하였다. 추출 용매는 methanol, ethanol 및 acetone 세 가지로, 각각의 추출 용매는 100%, 70% 그리고 70% 용매에 0.5% HCl을 포함한 세 가지 형태의 용매를 사용, 총 9가지 추출 용매로 실험을 수행하였다. 보관된 실험 재료는 추출 전 액체질소를 이용하여 분쇄하고 15 mL plastic tube에 500 mg 씩 정량하여 사용하였다. 정량된 과피 및 종자 샘플에 추출 용매 5 mL를 첨가한 후, 초음파 추출기(DH. EUC. D10H, 200W, 40KHz, Daehan Scientific)를 이용하여 30분 추출한 후, 원심분리기(VS-550, Vision Scientific)을 이용하여 1,500 rpm에서 10분간 원심 분리를 수행하였다. 원심 분리 후, 상등액은 새로운 15 mL plastic tube에 옮겼다. 남아있는 침전물에 추출용매 1 mL를 사용하여 추출을 한번 더 수행하였다. 추출된 상등액은 총 탄닌 함량 및 개별 탄닌을 분석하는데 사용되었다.

총 탄닌 함량 분석

포도의 총 탄닌 함량 분석은 Slingleton(16)의 방법을 참조하여 수행하였다. 추출된 상등액 20 μL와 증류수 1200 μL를 2 mL microtube에 넣고, 100% Folin-Ciocalteu phenol reagent 시약 100 μL 및 15% Na2CO3(Sodium carbonate)시약 300 μL를 순서대로 첨가하였다. 반응 시약을 첨가한 후, 최종 볼륨이 2 mL가 되도록 증류수 380 μL를 추가로 첨가하였다. 또한, 암조건의 실온에서 2시간 동안의 반응 시간을 거친 후, microplate로 200 μL씩 옮겨 다기능 흡광 분석기(Multiskan Go, Thermo Fisher Scientific, Waltham, USA)를 사용하여 765 nm에서 흡광도를 측정하였다. 총 탄닌 함량은 표준 시약 tannin acid를 사용하여 검량선을 작성하였으며, 모든 실험은 4반복으로 이루어졌다.

HPLC 분석

추출된 상등액은 HPLC 분석을 위해 추가적인 탄닌 분리 실험을 수행하였다. 총 탄닌 분석에 사용된 20 μL를 제외한 추출된 6 mL의 상등액을 진공 회전농축기(concentrator plus, Eppendorf, Hamburg, Germany)를 이용하여 용매층을 농축하고 최종 볼륨 3 mL가 되도록 H2O를 첨가하였다. 추출물에서 지방족 화합물을 제거하기 위하여 chloroform 5 mL를 첨가하고 5분간 초음파 추출기를 이용하여 혼합한 후, 1,500 rpm에서 10분간 원심 분리하였다. 상층용매(H2O층) 2 mL를 microtube에 옮겨 15,000 rpm에서 10분간 원심 분리하고 최종적으로 1 mL의 H2O층을 취하였다. 1 mL의 H2O층은 진공 회전농축기를 이용하여 30℃ 온도에서 완전 농축하고, 500 μL의 methanol으로 재용해 후, 0.22 μm의 membrane filter(Millipore, Burlington, USA)로 여과하여 HPLC 분석에 사용하였다.

DAD(Diode Array Detector)와 Capcell Pak C18 Column (4.6×250 mm I.D., Osaka Soda, Japan)이 장착된 HPLC (Agilent 1100 series, Agilent Technology, California, USA)을 이용하여 monomer 탄닌(catechin, epicatechin, gallocatechin, epigallocatechin) 및 dimer 탄닌(procyanidin B1, procyanidin B2)을 분석하였다. 주입량은 10 μL로 하였으며, 이동상은 용매 A(0.1% acetic acid in H2O)와 용매 B(100% acetonitrile)를 A:B 용매 혼합비율 90:10(1 min), 78:22(21 min), 50:50(23 min), 10:90(25 min), 10:90(28 min), 90:10(30 min), 90:10(33 min)의 조건으로 1 mL/min의 유속에서 분석하였다. 검출 파장은 280 nm이었으며 모든 분석은 4반복으로 이루어졌다.

통계분석

모든 실험구는 4반복으로 이루어졌으며 실험 결과의 통계 처리는 SAS 프로그램(SAS institute, Cary, NC, USA)를 사용하여 분산분석(ANOVA)을 하였으며, 처리간 유의차 검증은 Duncan’s multiple range test를 사용하여 95% 수준에서 검증하였다.

결과 및 고찰

총 탄닌 함량

Campbell Early 포도의 과피 및 종자에서 총 탄닌 함량을 측정한 결과, 과피에서는 0.5% HCl를 첨가한 70% acetone과 70% acetone 순으로 총 탄닌 함량이 높게 나타났다(Table 1). 그 외에 0.5% HCl를 첨가한 70% methanol, ethanol 추출 용매에서도 0.5% HCl를 첨가하지 않은 추출 용매에서보다 대부분 높은 수준의 탄닌이 추출되었다.

종자에서는 70% acetone과 0.5% HCl 첨가 70% acetone에서 탄닌 함량이 가장 높게 나타났다(Table 1). 가장 낮은 추출을 보인 용매 조건은 100% ethanol이었다. 종자의 탄닌 함량은 과피에서 보다 수배 이상 높게 확인되었으며 이는 포도 발달의 모든 단계에서 항상 종자에서 과피보다 높은 총 탄닌 함량을 나타내었다는 연구 결과와 유사하였다(17,18).

과피와 종자 모두 0.5% HCl를 첨가한 70% acetone에서 가장 높은 탄닌 추출을 보였으나 이는 0.5% HCl 첨가로 인해 탄닌 및 안토시아닌을 포함하는 폴리페놀이 함께 추출된 결과로 추정되었다. Novak 등(19)에 따르면 추출 용매에 산(acids)을 첨가하면 식물체의 분해를 촉진하여 세포질로부터 폴리페놀의 추출을 향상시킬 수 있다고 하였는데, 본 실험에서도 선행 연구와 같이 0.5% HCl를 첨가한 70% acetone 조건에서 가장 높은 탄닌 추출을 보였다. 그러나, 본 실험에서 확인한 총 탄닌 함량은 UV-spectrophotometer를 통하여 이루어졌으며, 같은 추출 방법으로 총 안토시아닌 및 총 폴리페놀 분석이 이루어지는 점을 감안할 때, 추출 시 탄닌, 안토시아닌 및 폴리페놀 함께 포함되어 있어 0.5% HCl를 첨가한 70% acetone 조건에서는 정확한 탄닌 측정이 이루어지지 않았을 것으로 추정되었다. 따라서 과피 및 종자의 총 탄닌 측정에 가장 적합한 추출 용매는 70% acetone으로 판단하였다.

특히, 총 탄닌 함량 분석은 포도 및 포도주의 떫고 씁쓸한 맛을 확인하기 위한 측정 방법으로 간편하여 적정 함량이 구명된다면 개별 탄닌 분석을 대체 가능할 것으로 판단된다(17). 그러나, 떫고 씁쓸한 맛을 나타내는 탄닌은 각기 다른 물질의 형태로 포도 및 포도주에 존재하므로 개별 탄닌의 분석도 필요하다 하겠다. 개별 탄닌의 측정은 HPLC, HPLC-DAD, 및 LC-MS/MS등의 실험 장비가 필요하며, 전처리의 간편화, 비용의 절감 및 실험의 용이성에서 HPLC가 가장 널리 사용되고 있다(19,20).

과피 및 종자의 탄닌 조성

포도의 떫은 맛과 씁쓸한 맛에 관여하는 것으로 알려진 monomer 및 dimer 탄닌을 HPLC-DAD를 사용하여 정량 분석하였다. 과피에서의 개별 탄닌 추출은 70% acetone과 70% ethanol에서 가장 높게 나타났다(Table 2). 포도 과피에서 gallocatechin, epigallocatechin 및 epigallocatechin gallate 등의 gallotannin이 주요 물질로 보고된 바 있으며(21,22), 본 연구에서도 주요 성분으로 gallocatechin 및 monomer 탄닌인 catechin과 dimer 탄닌인 procyanidin B1이 70% acetone 용매 조건에서 가장 높게 검출되어 과피의 탄닌 분석에 적합한 용매 조건으로 판단되었다.

종자에서의 개별 탄닌 추출은 70% acetone, 70% methanol 순으로 높게 나타났으나 두 처리 간 유의적 차이는 나타나지 않았다(Table 3). 종자의 주요 탄닌 성분으로 보고된(23,24), catechin, epicatechin, gallocatechin, epigallocatechin은 70% acetone 추출 용매에서 가장 높게 검출되었고, procyanidin B1은 70% methanol, procyanidin B2은 100% methanol 에서 각각 높게 검출되었다. Procyanidin B1, B2는 epicatechin에 각각 catechin과 epicatechin이 탄소 결합한 dimer 탄닌으로 monomer 탄닌과 더불어 포도 및 포도주의 씁쓸함(bitterness)과 연관되어 있는 것으로 보고되었다(1). 또한, methanol에서 가장 높은 추출을 보인 procyanidin B1, B2를 제외하고, 개별 탄닌은 수용성 추출 용매에서 높은 추출 함량을 나타내었다. 이는 Omar 등(25)의 결과와 유사한 것으로, 탄닌 추출 시 H2O 첨가는 100% 용매보다 높은 폴리페놀 추출을 나타냈다.

위의 결과에서 추출된 개별 탄닌이 과피와 종자의 용매 조건이 다름을 확인하였다. 과피에서는 70% acetone에서 gallocatechin와 procyanidin B1을 제외한 나머지 탄닌 성분은 70% methanol에서의 검출량과 유의차가 없었으며, 총 검출량에서도 가장 높은 값을 나타내어 가장 적합한 추출 용매로 판단하였다. 종자에서는 70% acetone과 70% methanol 추출 용매 조건에서 종자의 주요 탄닌 성분인 catechin과 epicatechin의 검출량이 높게 나타났으나, dimer 탄닌 procyanidin B1 성분이 70% methanol에서 높게 검출되어, 종자에서 개별 탄닌을 고루 검출하기 위해서는 70% methanol 추출 용매가 적합한 것으로 판단하였다

따라서 과피의 총 탄닌 함량과 개별 탄닌 조성 분석에는 70% acetone을 추출 용매로 사용하고 종자의 경우에는 총 탄닌 함량 분석에 70% acetone을, 개별 탄닌 추출에는 70% methanol을 사용하는 것이 적합한 것으로 판단되었다. 개별 탄닌의 추출 및 검출은 추출 방법과 추출 용매의 선택에 따라 다른 결과를 나타내며, 사용 분석 기기에 따라서도 추출 방법과 용매가 각기 다르다. 따라서 이러한 조건을 감안한 보편적이고 효율적인 추출 방법에 대한 연구는 계속되어야 할 것으로 판단된다.

요 약

본 연구에서는 포도 과피 및 종자에서 초음파 추출법을 통해 methanol, ethanol, acetone용매를 대상으로 각각 100%, 70%, 70% 용매에 0.5% HCl을 첨가한 조건에서 총 탄닌 함량 및 monomer와 dimer 탄닌 함량을 측정하였다. 과피의 총 탄닌 함량은 추출 용매 조건에 따라 0.01-2.92 mg/g으로 나타났으며 종자의 총 탄닌 함량은 8.82-14.72 mg/g 으로 나타났다. 그 결과, 총 탄닌 함량은 과피 및 종자에서 모두 70% acetone 조건에서 가장 우수한 검출량을 나타냈다. Monomer 및 dimer 탄닌 등 개별 탄닌 함량을 측정한 결과, 과피에서는 70% acetone이 가장 적합한 용매 조건으로 판단되었다. 종자에서는 70% acetone이 가장 높은 탄닌 검출량을 나타내었으나, monomer 탄닌 검출량이 70% methanol 조건과 유의차가 없었으며 dimer 탄닌이 70% methanol에서 높게 검출되어 70% methanol을 가장 적합한 용매 조건으로 판단하였다. 개별 탄닌에 대한 분석은 지금도 꾸준히 진행되고 있으며 분석 효율을 높이는 분석 방법에 대한 연구가 계속되어야 할 것으로 판단된다.
